Cannot connect to MySQL server (10061)

I am currently supporting a PHP website using MySQL.
When connecting to the MySQL database whilst on the server with:
mysql -h localhost -u USERNAME -p
We have no problems as it connects fine.
However when trying to connect from another machine on the LAN we get the error as shown in the topic so we cannot connect.
Please note that we use either the IP address or the host name instead of localhost for this.
We have no firewall running on the server and yet it is still blocking our activity.
I have opened port 3006 and allowed the mysql.exe file through the client firewall and can access other servers using the MySQL command.
Can anyone help me solve this problem as it is causing some issues whilst developing.
