Does the file documented by HKLM/Software/Microsoft/ODBC/ODBCINST.INI in the registry exist? If not, you may need to re-install the driver, as the error message suggests.

The MySQL ODBC driver couldn't know anything about Simply Accounting's requirements. Best ask the makers of SA.
