The rule of thumb would suggest more like 6-8GB for key_buffer_size on a 32GB machine. 12GB probably would not be too bad. Note: The two main uses for RAM (in a MyISAM-only setup) are the key buffer (for MyISAM indexes) and the OS's cache space (for MyISAM caching). Since your data and index sizes are much bigger than them, you will mostly be getting cache misses.
OTOH, if you have good "locality" in your accesses, you might not have that much disk activity.
What kind info do you have in the table(s)? If it is Data Warehouse-type application, there are other suggestions I could give.
