Creating an Index

I have a read mostly write rarely data file with about 2,000 entries that gets accessed up to 400,000 times a day. I want to speed up access by indexing but don't know too much about it. The records are not very large, total storage about 800K.

Access typically selects from 1 to 10 records from one column that have a common data value so that parameter would be the one to index. When I try to set it as a primary index it gets rejected, I assume because there is more than one record with the same index value.

In order to create an index I would have to add a second column of discrete data but data that is virtually never needed.

Does an index like this provide any advantage in accessing speed?
When I create it, the index occupies about 125K or 16% the size of the whole data file.
