Dozens, maybe hundreds of compatibility issues occur from one major version to the next. You are upgrading 2 versions.
On the other hand, there is only a slight chance that any of them will bite you.
Plan A: Wade through the changelogs. (Very tedious.)
Plan B: Set up a test, preferably on a separate machine. Practice the upgrade (there could be hiccups). Test the resulting 5.5.
Plan C: Make 5.5 a Slave to your 5.0 (Master). Test read operations on the Slave. Then failover.
Plan D: Update in-place. This has the least effort, but maximal risk.
