Yes, this is possible to run virtual machine with direct access to the real hard disk. You have to use, for example, Vmware Workstation (as far as I know 6.5+ versions can do this for sure, unsure about the earlier). Vmware Server (that is freeware) is unable to do this.
If you have the fast network and no Vmware Workstation you can use SAN solution (Such as StarWind) and mount the remote disk drive to machine like it was local one. For this task you need at least Gigabit Ethernet.
