It definitely works. You just have to get some connections going to it. If you have a high-traffic site, it will happen naturally. During testing I wrote a primer script to get a bunch of connections going.
Now I do hundreds of millions of queries per month through mysql-proxy and its definitely splitting up my read and r/w traffic. Cacti and dbmon.pl assure me of that : )
I'm working on some lua mods to split session DB out to a separate pool and not send ANY non-transaction selects to the primary rw server.
