In general, MySQL does a good job of handling such. I would guess that only 1-2% of DBAs have much trouble with such.

You could manually layout what goes on what drive, only to find that one table is running out of space or a temp table is hit the limit -- while there is lots of space elsewhere. Get the gun, aim for your foot, pull the trigger.
