I've asked a question in the InnoDB section which may clarify its limits. I've personally used more than 6GB of RAM for InnoDB on a Wikipedia server. Not a problem at that level (on suitable equipment, of course - dual Opteron and Fedora in that case).

I don't have a definitive answer on combined limits for the MySQL buffers. Judging from the manual it appears not to be a problem to go to very large combined sizes if you can divide your indexes into different key caches.
