If you have only InnoDB tables (no MyISAM), look at XtraBackup.
In general, you should look at Replication -- a Slave can serve as a backup or be a tool for performing a backup without impacting the Master. If you currently have a single server (no Replication), there will have to take a backup of that server in order to make it a Master and to initially populate the Slave.
Read the documentation online.
