Answering your questions:

1. No there's no real difference other than the distance MySQL network packets need to travel.

2. Yes, it will work as long as MySQL server has a 'user'@'host-or-ip' that matches the user name and the client hostname or IP address. Check documentation for CREATE USER for details https://dev.mysql.com/doc/refman/8.0/en/create-user.html
