Yes, INDEX(col1,col6,col10,col20,col13) would work well for both queries. And it is better to have one index, not two.

However, long indexes get to the point of "diminishing returns". 5 columns is about the limit of usefulness.

Also, it is not good to have more than, say, a dozen indexes.
