currently mysqlbackup can not do remote backups, so answering your first question, you must have MEB on server B.
Regarding to second question both solutions have advantages and disadvantages.
When you backup a slave you can be sure no slowdown occurs on master due to copying activity or short period of locking (if you backup non-InnoDB tables).
At the same time backing up the master you are sure you backed up latest changes. For slave always will be slight delay. But this delay can be OK if you backup binary logs from the master.
Actually backing up the slave and saving binary logs from master is recommended solution in most cases.
Another reason to backup master is if you use binary log or replication filters (binlog-* and replicate-* rules). If you have backing up the master is only option.
