Hi,

Do your queries contain join of very many tables? If not, changing optimizer_search_depth will not help. If yes, which MySQL version are you using?
MySQL 5.6 contains improvements in this area. (See http://jorgenloland.blogspot.co.uk/2012/04/improvements-for-many-table-joins-in.html)

Another source of long optimization times in earlier versions have been long IN lists. This is also improved in MySQL 5.6.
